SS-31 selectively partitions to the inner mitochondrial membrane (IMM) in a membrane potential-dependent manner, achieving local concentrations 1,000–5,000-fold greater than extracellular levels within minutes of exposure. Once localized to the IMM, SS-31 binds with high affinity to cardiolipin — a phospholipid unique to the mitochondrial inner membrane that serves as a structural and functional anchor for electron transport chain (ETC) complexes and cytochrome c. By stabilizing cardiolipin against peroxidative damage from reactive oxygen species (ROS), SS-31 preserves the quaternary organization of ETC supercomplexes, maintains cristae architecture, and prevents cytochrome c release into the intermembrane space — a key initiating event in mitochondrial apoptosis. SS-31 also reduces mitochondrial ROS production by improving electron flow efficiency through Complexes I, III, and IV, without functioning as a direct free radical scavenger. This cardiolipin-stabilizing mechanism distinguishes SS-31 from conventional antioxidants and enables targeted organelle-level cytoprotection.
